Stallergenes Greer announces publication of positive results for Palforzia® phase III study in peanut-allergic children aged 1 to 3 years

  • Full POSEIDON (Peanut Oral Immunotherapy Study of Early Intervention for Desensitization) study data published in New England Journal of Medicine Evidence.
  • 73.5% of patients treated with PALFORZIA met the primary endpoint of tolerating the 600 mg single dose of peanut protein at 12 months compared with 6.3% of patients receiving placebo.
  • No serious or severe treatment-related adverse events were reported with the use of PALFORZIA. Only 2% of patients experienced systemic allergic reactions related to the trial treatment.
  • Evidence of progression of peanut allergy in patients receiving placebo but not in patients treated with PALFORZIA.  

Baar, SwitzerlandNovember 20, 2023 – Stallergenes Greer, a leading global healthcare company specialising in allergen immunotherapy (AIT), announces the publication of results from the Phase 3 POSEIDON (Peanut Oral Immunotherapy Study of Early Intervention for Desensitization) study in the New England Journal of Medicine Evidence. The study evaluated the efficacy and safety of PALFORZIA [defatted powder of Arachis hypogaea L., semen (peanuts)] in peanut-allergic children aged 1 to 3 years.

The study met the primary endpoint, with the majority of patients treated with PALFORZIA tolerating at least a 600 mg single dose (1,043 mg cumulative) or a 1,000 mg single dose (2,043 mg cumulative) of peanut protein (73.5% and 68.4%, respectively, compared to 6.3% and 4.2% of patients who received placebo, respectively). In addition, the majority of patients treated with PALFORZIA (61.2% compared with 2.1% of placebo-treated patients) tolerated the highest exit double-blind, placebo-controlled exit food challenge (DBPCFC) dose level of 2,000 mg (4,043 mg cumulative), and this proportion was higher for the youngest age group.

PALFORZIA demonstrated a favorable safety profile. Overall, 84.7% of patients on PALFORZIA and 93.8% of patients on placebo completed the study. There were no PALFORZIA-related serious or severe adverse events, and systemic allergic reactions due to any cause were similar between PALFORZIA and placebo-treated patients (8.2% and 8.3%, respectively). Treatment-related systemic allergic reactions occurred in 2% of PALFORZIA-treated patients and 0% of placebo-treated patients.

A 3.5-fold increase in peanut allergy prevalence has been reported in recent years, with 1 to 2% of children in Western countries affected1. Reactions to peanut are potentially life-threatening, accounting for the majority of deaths related to food allergy2.  

Through the Poseidon study, we have witnessed a significant shift in how peanut allergy can be managed in the youngest patients,” said George du Toit, M.B., B.Ch., Professor of Pediatric Allergy at Evelina London Children’s Hospital, Guy's and St Thomas' NHS Foundation Trust, King’s College London and study investigator in PALFORZIA clinical trials. “These findings highlight the potential benefit of early intervention for peanut-allergic children aged 1 to 3 years, offering a glimpse of hope for a future where we can alleviate the burden of this condition.”

ABOUT POSEIDON PHASE 3 STUDY

POSEIDON (Peanut Oral Immunotherapy Study of Early Intervention for Desensitization, clinicaltrials.gov number NCT03736447) is an international, randomized (2:1), double-blind, placebo-controlled Phase 3 study that evaluated the efficacy and safety of PALFORZIA in peanut-allergic children aged 1 to <4 years of age in North America and Europe. The POSEIDON study was completed by Aimmune Therapeutics, part of Nestlé Health Science before Nestlé divested PALFORZIA to Stallergenes Greer in September 2023. Enrollment was based on several entry criteria, including a documented clinical history of peanut allergy, positive skin prick tests and/or elevated blood levels of peanut antibodies, and dose-limiting symptoms after consuming single doses of peanut protein >3 to ≤300 mg in a positive double-blind, placebo-controlled food challenge (DBPCFC). In POSEIDON, patients underwent a dose-escalation period of approximately 22 weeks to reach a dose of 300 mg per day of PALFORZIA or placebo, then continued that dose for approximately six months. At the end of the trial, patients underwent an exit DBPCFC.

ABOUT PALFORZIA 

PALFORZIA is a complex biologic drug used with a structured dosing approach that builds on a century of oral immunotherapy (OIT) research. With OIT, the specific allergenic proteins are ingested initially in very small quantities, followed by incrementally increasing amounts, that can result in the ability to mitigate allergic reactions to the allergen over time. PALFORZIA is a rigorously developed, pharmaceutical-grade OIT for peanut allergy with a well-defined allergen profile to assure the consistency of every dose, from 0.5 mg (equivalent to 1/600th of a peanut) to 300 mg.

PALFORZIA is not intended for, and does not provide, immediate relief of allergic symptoms. Therefore, this medicinal product is not to be used for emergency treatment of allergic reactions, including anaphylaxis. Self-injectable adrenaline (epinephrine) must be available to the patient at all times.

PALFORZIA is approved by the U.S. Food and Drug Administration (FDA) by the European Medicine Agency (EMA), by the Medicines and Healthcare products Regulatory Agency (MHRA) in the U.K., and by Swissmedic in Switzerland.

Stallergenes Greer announces global agreement with Nestlé regarding the peanut allergy oral immunotherapy Palforzia®

Baar (Switzerland), September 4, 2023 – Stallergenes Greer, a leading global healthcare company specialising in allergen immunotherapy (AIT), announced today that it has signed global agreements with Nestlé to harness the peanut allergy oral immunotherapy treatment Palforzia®. The transaction was closed upon signing. 

Peanut allergy currently affects approximately 2% of the general population of Western nations1, and the prevalence of peanut allergy doubled among children between 2005 and 20152. The difficulty of avoiding peanuts, combined with the severity of allergic reactions, including anaphylaxis, shows the need for effective treatment3

The company’s portfolio offers patients and healthcare professionals precise personalised AIT treatments which cover a broad range of administration modes and can be tailored to the individual needs of patients with allergies: sublingual oral solutions, tablets, and injectables for respiratory allergies; and powder to be consumed with semi-solid foods for peanut allergies.  

 

ABOUT PALFORZIA® 

Palforzia® is an oral immunotherapy treatment approved for the mitigation of allergic reactions, including anaphylaxis, that may occur with accidental exposure to peanut for patients aged 4 to 17 years with a confirmed diagnosis of peanut allergy by the U.S. Food and Drug Administration (FDA), the European Commission (EC), the Medicines and Healthcare products Regulatory Agency (MHRA) in the U.K., and by Swissmedic. Use of Palforzia® may be continued in patients 18 years of age and older.

ABOUT STALLERGENES GREER STUDIES EfficAPSI AND PRACTIS

EfficAPSI is the largest retrospective real-world, longitudinal cohort study regarding sublingual liquid allergen immunotherapy treatment. Its objective is to evaluate the real-life impact of sublingual liquid allergen immunotherapy on the onset and worsening of asthma in patients with allergic rhinitis. This study included more than 100,000 patients in France with allergic rhinitis with or without asthma treated with sublingual liquid AIT and symptomatic drugs and more than 330,000 patients with allergic rhinitis with or without asthma treated with symptomatic drugs only.

PRACTIS is a French multicentre real-world observational prospective one-year study. It evaluates over 1,000 allergic children, adolescents and adults treated in current practice with sublingual immunotherapy (tablets and sublingual solutions) to analyse how SLIT treatment meets patient expectations through the patient benefit index.

Stallergenes Greer Foundation Science Awards for Allergy 2023: Call for applications

Baar (Switzerland), June 8, 2023 – The Stallergenes Greer Foundation aims to create healthier futures for all. The Foundation, under the aegis of the Fondation de France, is philanthropically dedicated to three focus areas:

  • Advancing scientific research to bolster innovation and precision medicine.
  • Supporting academic initiatives to further develop future generations of allergy healthcare professionals.
  • Engaging in climate action and environmental protection to progress the prevention and treatment of allergies.

Submissions are open for the Stallergenes Greer Foundation Science Awards for Allergy 2023. A total of €150,000 will be awarded for outstanding work led either by a young scientist or a mid-career scientist to advance precision medicine in the field of allergy, or outstanding work in the environmental field to advance the prevention and treatment of allergies.

The Stallergenes Greer Foundation Science Awards for Allergy are presented in two areas:

- Innovation Awards: two awards will be presented either to encourage and raise the profile of a young scientist (below 40 years of age) or to recognise the outstanding work of a mid-career scientist (with a 10–20-year track record), who are driving innovation in respiratory and food allergy.

- Environmental Health Award: the award will honour the work of a researcher who is pioneering research aimed at preserving and restoring both natural and built ecosystems and contributing to advancing the prevention and treatment of allergies.

Timeline: The submission period is open from July 1, 2023, to November 29, 2023, at 11:59pm CET. Submissions will be reviewed by the Stallergenes Greer Foundation’s Board of Trustees. Decision letters will be sent out at the end of February 2024.

Eligibility: the awards are open to physicians, PhDs and Pharm-Ds actively engaged in allergy research and attached to a research team in a non-profit research laboratory. There are no geographic limitations.

Stallergenes Greer initiates a European paediatric Phase III Study with STALORAL® Birch

Baar (Switzerland), May 25, 2023 Stallergenes Greer, a biopharmaceutical company specialising in the diagnosis and treatment of respiratory allergies, announced today the initiation of a Phase III clinical study (YOBI, YOung patients and BIrch allergy) to confirm the safety and efficacy of Staloral® Birch in children and adolescents with birch pollen-induced allergic rhino-conjunctivitis with or without asthma. The randomised, double-blind placebo-controlled Phase III study will involve approximately 80 sites in 12 European countries.

Allergic rhino-conjunctivitis is a worldwide disease and a global health burden affecting more than 500 million people, who are at higher risk of developing rhinitis exacerbation and asthma than the general population[1]..Birch is among the strongest allergy-producing trees in northern Europe. The allergic potential of other trees belonging to the birch homologous group, such as alder and hazel, is also increasing steadily[2].

In children and adolescents, allergic rhino-conjunctivitis can lead to sleep problems, fatigue, missed school days and can complicate outdoor activities[3]. Individuals sensitised at a young age are also more at risk of developing asthma. The younger the children are, the higher their risk of developing asthma, which highlights the relevance of early causal treatment in paediatric patients to slow the worsening of allergy symptoms[4]. Allergen immunotherapy (AIT) is currently the only available therapeutic treatment that has the potential to uniquely alter the natural course of the disease[5]. In its liquid form, sublingual AIT can provide a needle-free and flexible treatment for children and adolescents.

Allergic rhino-conjunctivitis induced by birch and related trees can lead to a spiraling of respiratory symptoms and conditions in the paediatric population. The YOBI study will contribute to addressing a global health burden, while deepening our knowledge of AIT with the aim of improving patient outcomes for children and adolescents with birch-induced allergies,” said Professor Oliver Pfaar, International Coordinator of the YOBI study, Head of Rhinology and Allergy, Department of Otorhinolaryngology, Philipps-Universität Marburg, Germany.

ABOUT STALORAL®

Staloral® is a sublingual solution of allergen extracts for AIT. It is indicated in the treatment of seasonal or perennial allergic rhinitis, rhino-conjunctivitis and mild to moderate allergic asthma in adults and, in certain territories, in the paediatric population (children over 5 years of age).

Clinical evidence of the efficacy of Staloral® is globally consistent over the clinical studies Stallergenes Greer conducted versus placebo. ABOUT ALLERGIC RHINITIS/ALLERGIC RHINO-CONJUNCTIVITIS

Allergic rhinitis/allergic rhino-conjunctivitis (AR/ARC) is a chronic disorder of the upper airways that is caused by allergen exposure and the resulting IgE-mediated inflammation of the nose and, to a less extent, the eyes. Allergic rhinitis can notably include symptoms such as sneezing, a runny nose, wheezing, cough, itching, and watery/itchy eyes. Symptoms may be severe and significantly impact the patient’s quality of life, as well as worsen over time with progression towards asthma4.

First intention symptomatic treatments such as antihistamines and nasal corticosteroids provide temporary relief to patients. The primary therapeutic goals of AIT include reducing symptoms, reducing symptomatic medication use, and improving allergy-related quality of life. AIT achieves these goals and can alter the course of the disease, with benefits persisting in many patients for several years after treatment discontinuation[6].

In 2023, the foundation will allocate up to €150,000 for outstanding work led either by a young scientist or a mid-career scientist to advance precision medicine in the field of allergy, or outstanding work in the environmental field to advance the prevention and treatment of allergies.

The Stallergenes Greer Foundation is governed by a Board of Trustees, which itself brings together a Managing Board and a Scientific Board:
